The application of hyperspectral remote sensing to coast environment investigation

Abstract:
Requirements for monitoring the coastal zone environment are first summarized. Then the application of hyperspectral remote sensing to coast environment investigation is introduced, such as the classification of coast beaches and bottom matter, target recognition, mine detection, oil spill identification and ocean color remote sensing. Finally, what is needed to follow on in application of hyperspectral remote sensing to coast environment is recommended.
Analysis of the high and low temperature centers and their relation with circulations in the East China Sea

Abstract:
Three warm currents, the Kuroshio, its shelf intrusion branch in the northeast of Taiwan and the Taiwan Warm Current (hereafter TWC), dominate the circulation pattern in the East China Sea (hereafter ECS). Their origination, routes and variation in winter and summer are studied. Their relationship with four major high and low temperature centers is analyzed. Differing from the previous opinion, we suggest that the four major centers are generated to a great extent by the interaction of the currents in the ECS. In summer, a cold water belt in the northeast of Taiwan is preserved from winter between the Kuroshio and the TWC. The shelf intrusion branch of the Kuroshio separates the water belt, and two low temperature centers generate in the northeast of Taiwan. In the southern ECS, the TWC transports more heat flux northward to form a warm pool. But it is separated in the lower layer by the cold water driven by the intrusion branch of the Kuroshio. So the TWC and the intrusion branch of the Kuroshio play a dominating role to generate the high temperature center. The interaction among the eastward TWC, the northward Tsushima Warm Current (hereafter TSWC) and the southward Su Bei Coastal Flow (hereafter SBCF) generates the low temperature center in the northern ECS. In winter, the strengthening of the shelf intrusion branch of the Kuroshio obscures the two low temperature centers in the northeast of Taiwan. For the weakening of the TWC, the high temperature center in the southern ECS vanishes, and the low temperature center in the northern ECS shifts to south.
Detection of nighttime sea fog/stratus over the Huanghai Sea using MTSAT-1R IR data

Abstract:
A dual channel difference (DCD) method is applied to detect nighttime sea fog/stratus over the Huanghai Sea using the infrared (IR) data of shortwave (3.5-4.0 μm) and longwave (10.3-11.3 μm) channels from the Multi-functional Transport Satellite (MTSAT)-1R, i.e., shortwave minus longwave brightness temperature difference (SLTD). Twenty-four sea fog events over the Huanghai Sea during March to July of 2006 and 2007 are chosen to determine a suitable value of SLTD for nighttime sea fog/stratus detection, and it is found that the value of -5.5-2.5℃ can be taken as a criterion. Two case examples of sea fog events are especially demonstrated in detail utilizing the criterion, and the results show that the derived sea fog/stratus coverage is quite reasonable. This coverage information is very helpful to analyze the formation and evolution of sea fog/stratus during night and can provide sea fog researchers with observational evidences for model results verification. However, more efforts are needed to further obtain vertical extent information of sea fog/stratus and attempt to discriminate between sea fog and stratus.
Reconstruction of incomplete satellite SST data sets based on EOF method

Abstract:
As for the satellite remote sensing data obtained by the visible and infrared bands inversion, the clouds coverage in the sky over the ocean often results in missing data of inversion products on a large scale, and thin clouds difficult to be detected would cause the data of the inversion products to be abnormal. Alvera et al.(2005) proposed a method for the reconstruction of missing data based on an Empirical Orthogonal Functions (EOF) decomposition, but his method couldn't process these images presenting extreme cloud coverage(more than 95%), and required a long time for reconstruction. Besides, the abnormal data in the images had a great effect on the reconstruction result. Therefore, this paper tries to improve the study result. It has reconstructed missing data sets by twice applying EOF decomposition method. Firstly, the abnormity time has been detected by analyzing the temporal modes of EOF decomposition, and the abnormal data have been eliminated. Secondly, the data sets, excluding the abnormal data, are analyzed by using EOF decomposition, and then the temporal modes undergo a filtering process so as to enhance the ability of reconstructing the images which are of no or just a little data, by using EOF. At last, this method has been applied to a large data set, i.e. 43 Sea Surface Temperature (SST) satellite images of the Changjiang River (Yangtze River) estuary and its adjacent areas, and the total reconstruction root mean square error (RMSE) is 0.82℃. And it has been proved that this improved EOF reconstruction method is robust for reconstructing satellite missing data and unreliable data.
Influence of particle composition on thorium scavenging in the marginal China seas

Abstract:
Thorium-234 and particle composition (organic matter, biogenic silica, carbonate and lithogenic component) were examined in the East China Sea (ECS) and the northern South China Sea (NSCS) in order to constrain the particle types scavenging thorium isotopes. Good positive correlations between particulate organic matter (POM) or carbonate and thorium-234 in suspended particulate matter (SPM) indicates that POM and carbonate are efficient to scavenge thorium-234. No relationship between biogenic silica and thorium-234 suggests that geochemical behavior of thorium-234 may be not influenced by biogenic silica. A simple model was used to evaluate the affinity of thorium-234 to different particle components. The results show that POM is the most efficient scavenger for thorium-234 in the ECS and the NSCS, followed by carbonate. The authors' results lend support to the utility of thorium-234 as a proxy of POC and carbonate in the upper layer. However, the strong dependence of thorium scavenging on particle composition challenges thorium-230 as a constant flux proxy.
The decay kinetics of residual chlorine in cooling seawater simulation experiments

Abstract:
To find out the decay character of residual chlorine (RC) in the sea water, the concentration of RC was analyzed by N, N-diethyl-p-phenylenediamine (DPD) method under different simulation experimental conditions, in which salinity, temperature, and Chemical Oxygen Demand (COD) were selected. The water used in the experiment was the mixture of aging ocean water, coastal water and extracting solution of coastal sediment at appropriate level. Results are shown as follows:(1) Piecewise function can well reflect the decay dynamics of RC in the cooling seawater. Concretely, the decay dynamics of first 1 min is too rapid to ascertain using a specific kinetic function, and that of the time from 1 to 30 min is fit for the first-order kinetic model. (2) The results could be the foundation of the chemical behavior of RC in seawater, and be used as not only the guidance of the coastal power plants production and sea water desalting companies, but also the establishment of the correlative trade standard.
Multivariate analysis of fluorescence and source identification of dissolved organic matter in Jiaozhou Bay, China

Abstract:
Hierarchical clustering analysis and principal component analysis (PCA) methods were used to assess the similarities and dissimilarities of the entire Excitation-emission matrix spectroscopy (EEMs) data sets of samples collected from Jiaozhou Bay, China. The results demonstrate that multivariate analysis facilitates the complex data treatment and spectral sorting processes, and also enhances the probability to reveal otherwise hidden information concerning the chemical characteristics of the dissolved organic matter (DOM). The distribution of different water samples as revealed by multivariate results has been used to track the movement of DOM material in the study area, and the interpretation is supported by the results obtained from the numerical simulation model of substance tracing technique, which show that the substance discharged by Haibo River can be distributed in Jiaozhou Bay.
Seasonal variations of dimethylsulfide (DMS) and dimethylsulfoniopropionate (DMSP) in the sea-surface microlayer and subsurface water of Jiaozhou Bay and its adjacent area

Abstract:
The distributions of DMS and its precursor dimethylsulfoniopropionate, in both dissolved (DMSPd) and particulate fractions (DMSPp) were determined in the sea-surface microlayer and corresponding subsurface water of the Jiaozhou Bay, China and its adjacent area in May and August 2006. The concentrations of all these components showed a clear seasonal variation, with higher concentrations occurring in summer. This can be mainly attributed to the higher phytoplankton biomass observed in summer. Simultaneously, the enrichment extents of DMSPd and DMSPp in the microlayer also exhibited seasonal changes, with higher values in spring and lower ones in summer. Higher water temperature and stronger radiant intensity in summer can enhance their solubility and photochemical reaction in the microlayer water, reducing their enrichment factors (the ratio of concentration in the microlayer to that in the corresponding subsurface water). A statistically significant relationship was found between the microlayer and subsurface water concentrations of DMS, DMSP and chlorophyll a, demonstrating that the biogenic materials in the microlayer come primarily from the underlying water. Moreover, our data show that the concentrations of DMSPp and DMS were significantly correlated with the levels of chlorophyll a, indicating that phytoplankton biomass might play an important role in controlling the distributions of biogenic sulfurs in the study area. The ratios of DMS/chlorophyll a and DMSPp/chlorophyll a varied little from spring to summer, suggesting that there was no obvious change in the proportion of DMSP producers in the phytoplankton community. The mean sea-to-air flux of DMS from the study area was estimated to be 5.70 μmol/(m2·d), which highlights the effects of human impacts on DMS emission.
A study of the microbial mineralization in submarine black smoker chimneys from the Okinawa Trough

Abstract:
Large amounts of microfossil records discovered in the seafloor black smoker chimney are reported from the Okinawa Trough. They are well preserved and can be divided into four types of filamentous microfossils. It suggests that the fossils may be derived from sulfur or iron oxidation chemolithotrophic prokaryotes and fungi. Based on the comparison studies of the microbial mineralization processes, two steps of biomineralization were hypothesized:(1) biology controlled mineralization; and (2) biology induced mineralization. At the early stage of the mineralization, the biology controlling mineralization is dominating; at the later stage, the biology inducing mineralization is the main mechanism. The composition of the fluids and the species of the microbes will determine the types of the minerals formed.
Cryopreservation of amphioxus (Branchiostoma belcheri) spermatozoa

Abstract:
Amphioxus, Branchiostoma belcheri, a closest relative of vertebrates, is at a high risk of extinction due to a combination of low effective population size, altered native habitats and environmental pollution, yet little is known about cryopreservation of its gametes. This study deals with the cryopreservation of amphioxus semen. The main findings are that (1) the extender of Yao et al. is the best one among the four extenders examined; (2) the appropriate ratio of semen to extender of Yao et al. plus cryoprotectant is from 1:5 to 1:7; (3) dimethyl sulfoxide (DMSO) and methanol are the better cryoprotectants than glycerol, with DMSO giving the best results; (4) the eggs fertilized with post-thaw spermatozoa are capable of developing to at least hatching stage, and the highest hatching rate is (12.4±3.0)%. This is the first report on freezing and thawing of amphioxus spermatozoa, providing a simple and practical protocol for cryopreservation of amphioxus spermatozoa and laying a foundation for safeguarding this endangered species.
Identification of Prorocentrum minimum and Takayama pulchella by fluorescence in situ hybridization through epifluorescence microscopy and flow cytometry

Abstract:
Partial rDNA sequences of Prorocentrum minimum and Takayama pulchella were amplified, cloned and sequenced, and these sequence data were deposited in the GenBank. Eight oligonucleotide probes (DNA probes) were designed based on the sequence analysis. The probes were employed to detect and identify P. minimum and T. pulchella in unialgal and mixed algal samples with a fluorescence in situ hybridization method using flow cytometry. Epifluorescence micrographs showed that these specific probes labeled with fluorescein isothiocyanate entered the algal cells and bound to target sequences, and the fluorescence signal resulting from whole-cell hybridization varied from probe to probe. These DNA probes and the hybridization protocol we developed were specific and effective for P. minimum and T. pulchella, without any specific binding to other algal species. The hybridization efficiency of different probes specific to P. minimum was in the order:PM18S02 > PM28S02 > PM28S01 > PM18S01, and that of the probes specific to T. pulchella was TP18S02 > TP28S01 > TP28S02 > TP18S01. The different hybridization efficiency of the DNA probes could also be shown in the fluorescent signals between the labeled and unlabeled cells demonstrated using flow cytometry. The DNA probes PM18S02, PM28S02, TP18S02 and TP28S01, and the protocol, were also useful for the detection of algae in natural samples.
Influence of the earth rotation on the interfacial wave solutions and wave-induced tangential stress in a twolayer fluid system

Abstract:
Interfacial waves and wave-induced tangential stress are studied for geostrophic small amplitude waves of two-layer fluid with a top free surface and a flat bottom. The solutions were deduced from the general form of linear fluid dynamic equations of two-layer fluid under the f-plane approximation, and wave-induced tangential stress were estimated based on the solutions obtained. As expected, the solutions derived from the present work include as special cases those obtained by Sun et al. (2004. Science in China, Ser. D, 47(12):1147-1154) for geostrophic small amplitude surface wave solutions and wave-induced tangential stress if the density of the upper layer is much smaller than that of the lower layer. The results show that the interface and the surface will oscillate synchronously, and the influence of the earth's rotation both on the surface wave solutions and the interfacial wave solutions should be considered.

Detection of Prorocentrum donghaiense using sandwich hybridization integrated with nuclease protection assay

Abstract:
Prorocentrum donghaiense is an important harmful algae bloom (HAB) causing creature in China's seas, and the conventional visual detection can not cope with long-term monitoring and highthroughput sampling projects. An assay for P. donghaiense with sandwich hybridization integrated with nuclease protection assay (NPA-SH) was established. Tests with mixed samples and spiked field ones confirmed its good specificity and sensitivity. The cell number of P. donghaiense correlated well with the optical density, and the regression equation is y=4×10-6x+ 0.694 9, in which x is the cell number, and y is the optical density, with r2=0.953 5. These results show that the NPA-SH method has good feasibility in the detection of P. donghaiense. Results of NPA-SH and microscopy are excellent for each sample. The NPA-SH method was a simple way in quantitative detection of P. donghaiense, and the whole process could be finished in about six hours, which provided a new approach in high-throughput sampling and long-term monitoring of P. donghaiense.
